Kalbin zamanı, kuşakları aşan ve kilit noktaları üç farklı zaman diliminde oluşan bir aşk üçgenini konu alıyor. Filmin tüm hikâyesini, emekli bir dedektifin ağzından dinliyoruz: Bir arkadaşına, yirmi yıl önce tanık olduğu, aynı kadına aşık olan üç gençle ilgili bir olayı hatırlayan komser, gençlerden birinin ölümü ile sonuçlanan bu olay üzerinde bir süre çalıştıysa da olayı aydınlatamamış ve nihayetinde bu dosyanın gencin 'intihar' ettiği sonucuyla kapanmasını hiçbir zaman içine sindirememiştir. Bunun bir cinayet olduğuna inanan emekli dedektif aradan geçen yıllara aldırmadan yeniden katilin peşine düşecek ve bizi de yanına katarak zamanda bir yolculuğa çıkaracaktır.
Ali Özgentürk, gençlik, aşk, dostluk ve mizahla harmanladığı "Kalbin Zamanı"nı şu cümlelerle özetliyor: "Üç adam bir kadını sevdi.... Kadın da onları.... Aşk 50 yıl sürdü.... İçlerinden biri öldürüldü...."
